RNA editing in flowering plants alters 400-500 and 30-40 cytidines to uridine in mitochondria and plastids, respectively. PLS class pentatricopeptide repeat (PPR) proteins have been assigned to be involved in single or few RNA editing sites in plant organelles. Recently proposed “PPR code” in which nucleotide identity of each PPR module is determined by the amino acid combinations at two key positions basically explains the sequence specificity of each editing factor, though it requires further improvement. In contrast to the PPR proteins, the multiple organellar RNA editing factors (MORF) proteins influence editing at many sites, for several of which individual PPR proteins are also required. Selective interactions between MORF proteins and PPR proteins are necessary for the editing function, implying formation of complex RNA editosomes in plant organelles. I discuss the possible function of MORF proteins in the RNA editosomes with 3D structural data.
